چکیده

were either unresponsive or showed elevated mRNA levels of the Th1-promoting chemokines CXCL10 and CXCL11. Conclusions: This study supported by grant SFB-F1802 of the Austrian Science Fund shows for the first time that the allergen uptake is specific and similar in DCs from allergic and healthy individuals. More important, the ensuing signal cascade that is triggered by the allergen differs between DCs of the 2 donor groups and results in a Th2-polarized immune response in allergic individuals as compared to ignorance/tolerance in normal donor cells.

Basophil expression of diamine oxidase: a novel biomarker of allergen immunotherapy response.

BACKGROUND Immunotherapy inhibits basophil histamine release, but the assay is cumbersome, and no one has studied the effects of immunotherapy withdrawal. OBJECTIVE Intracellular fluorochrome-labeled diamine oxidase (DAO) was used as a novel functional readout of basophil histamine release after immunotherapy. Results were compared with conventional basophil surface expression of activation m...

Optimizing of the basophil activation test: Comparison of different basophil identification markers.

BACKGROUND Flowcytometric identification of basophils is a prerequisite for measuring activation of basophils with IgE-dependent or IgE-independent stimuli. Aim of this study was to compare different marker combinations in a simultaneous multicolor flowcytometric measurement. METHODS Ten patients with a grass pollen allergy and three controls were included in the study. Basophilic cells were ...

Basophil sensitivity through CD63 or CD203c is a functional measure for specific immunotherapy

BACKGROUND Subcutaneous Immunotherapy (SCIT) modifies the allergic response and relieves allergic symptoms. SCIT is the only and a very effective treatment for insect venom allergy. We hypothesized that basophil sensitivity, measured through the basophil activation test, would decrease during SCIT up dosing. Expression of CD203c was compared to CD63 as marker for basophil activation, using a Bl...

Basophil CD203c levels are increased at baseline and can be used to monitor omalizumab treatment in subjects with nut allergy.

RATIONALE Basophils contribute to anaphylaxis and allergies. We examined the utility of assessing basophil-associated surface antigens (CD11b/CD63/CD123/CD203c/CD294) in characterizing and monitoring subjects with nut allergy. METHODS We used flow cytometry to analyze basophils at baseline (without any activation) and after ex vivo stimulation of whole blood by addition of nut or other allerg...

Biomarkers for allergen immunotherapy in cedar pollinosis.

To initiate, monitor, and complete effective immunotherapy, biomarkers to predict and visualize the immune responses are needed. First, we need to identify the right candidate for immunotherapy. Secondly, the immune responses induced by immunotherapy should be monitored. For the first objective, analysis of polymorphisms of candidate genes may be helpful, but still be in development. Regarding ...
